生物分子间二维与三维反应动力学的比较研究 [PDF]

生物大分子间反应可分为束缚分子的二维反应和游离分子的三维反应。根据描述二维反应动力学的概率动力学模型,可以设计实验来测定分子间二维反应动力学,例如微管吸吮(Micropipette)、光镊(OpticalTrap)、原子力显微镜(AFM)等技术,但上述方法较复杂,一般只用于实验室研究。基于表面等离子体共振技术的生物传感器是成熟的、商业化测定三维反应动力学方法,该方法简单、易掌握,已经广泛应用于药物的设计、筛选和评价,但难以反映在体条件下大部分生理现象和生物学过程是由二维反应所介导的实际。
