【目的】应用基质辅助激光解吸电离飞行时间质谱技术检测人类载脂蛋白E(apoE)基因的多态性。【方法】用PCR扩增出含apoE两个单核苷酸多态性(SNPs)位点(334T/C和472C/T)的DNA片段作为模板,经延伸反应,产物纯化后利用MALDI-TOFMS进行检测。从而确定样本apoE基因型。【结果】应用本技术对50例广东地区无关个体进行apoE基因多态性检测,共检出3种常见的apoE基因型,其中以apoEΣ3/3型最多,占总数的74.00%;等位基因频率分布以ε3(87.00%)最高。【结论 ...
