利用DNA聚合酶进行DNA体外复制时,模板上的多种回文结构可能导致复制跳跃,表现为较大的片段缺失。本文在研究黑斑侧褶蛙抗菌肽Temporin基因结构时,对一个1126 bp的内含子片段进行PCR扩增、扩增产物的T-A克隆和Sanger测序,在这3个阶段都遇到了在同一位置出现564 bp的缺失。根据实验数据和文献报道,我们推测这一复制跳跃的产生与一段6 bp的同向重复单元有关,并提出跳跃可能发生的机制:模板上两个重复单位下相互靠近,形成特定的高级结构,使DNA聚合酶脱落下来,新链延伸被阻断 ...
