高原地区大鼠耳蜗RNA提取条件优化

目的探讨在高原地区采用AMBION抽提试剂盒提取RNA的最佳条件。方法采用AMBION抽提试剂盒,对126份大鼠耳蜗(每份耳蜗分为2组,实验组和对照组)提取RNA,实验组采用AMBION抽提试剂盒优化条件提取RNA,对照组按照AMBION抽提试剂盒说明书提取RNA。结果实验组和对照组均能成功提取大鼠耳蜗组织RNA,总RNA的提取量分别为:0.896±0.348μg,0.536±0.404μg(P>0.05);实验组RNA浓度高于对照组,分别为:0.147±0.047μg/μl,0.072±0.022μg/

石蜡包埋淋巴组织三种微量DNA提取方法的比较

目的为寻找一种高效、简便、实用的石蜡组织中DNA提取方法,本研究比较了三种不同的DNA提取方法对DNA质量的影响。方法选取淋巴相关组织的蜡块21例。三种DNA提取方法分别是简单消化煮沸法,酚/氯仿提纯法和试剂盒法。通过提取DNA的浓度和纯度、PCR扩增保守序列β-globin等方法,比较三种DNA提取方法的优劣。结果试剂盒法提取样品OD260/OD280平均比值位于1.61.8之间;提纯法位于1.31.5之间;简单消化煮沸法位于1.21.3之间。三种方法中,试剂盒法提取的DNA最为稳定 ...

不同提取方法对山桐子果实挥发油成分的影响

用水蒸气蒸馏法、超声辅助浸提法和索氏提取法提取山桐子果实中的挥发油,用GC-MS分离鉴定其成分并测定各成分的相对含量.水蒸气蒸馏法、超声辅助浸提法和索氏提取法分别分离鉴定出64、50和57个组分,共有化合物38种,其中主要成分为亚油酸、棕榈酸、油酸、角鲨烯、椰子醛和γ-维生素E等.3种提取方法所得山桐子果实挥发油成分有共同特征,也存在着差异,相较之下,水蒸气蒸馏法提取的挥发油成分最多,索氏提取法提取挥发油率最高.
