目的筛选与弱精子症发生相关的基因。方法采集正常男性和弱精患者精液精子,非连续梯度离心法分离纯化精子,收集95%percoll以下和57%与76%Percoll层之间的精子,以排除生精细胞和白细胞的污染。将纯化后精子的cDNA探针与Affymetrix全基因组芯片杂交。筛选出差异表达的基因,利用生物信息学方法对该基因进行分析。用RT-PCR验证该基因在正常男性和弱精患者精液精子中的差异表达,并用RT-PCR方法分析该基因在人不同组织(心、肝、脾、肺、肾、胃、脑、睾丸)中的表达。结果分析芯片杂交结果 ...

性激素对中枢甲硫氨酸脑啡肽含量的影响

用放免法测定大白鼠脑之纹状体、海马、下丘脑、垂体前叶与后叶中甲硫氨酸脑啡肽(metenkephalin,MEK)的含量,显示去势雌鼠纹状体、海马、下丘脑、垂体前叶MEK明显下降,去势雄鼠下丘脑、垂体前叶也明显下降,注射已烯雌酚,黄体酮及睾丸酮的雌鼠,下丘脑MEK明显降低而垂体后叶明显升高,注射睾丸酮的雄鼠,垂体前叶与后叶MEK下降,雄鼠注射已烯雌酚,未见有显著改变,提示性腺与性激素对中枢MEK含量有影响。
