Study on an amylase-producing actinomycetes from ocean: isolation, identification, mutation and optimization of culture conditions(海洋来源产淀粉酶放线菌的分离鉴定、诱变选育及培养条件优化)

采用鉴定培养基初步筛选产淀粉酶的菌株,利用YOO改良法测定其酶活力,复筛产酶能力强的菌株作为研究对象.通过热、超声波及紫外线等多种物理诱变选育高产淀粉酶菌株;采用单因子及正交试验优化菌株产酶培养条件.通过形态鉴定、生理生化鉴定以及分子生物学手段对其16S rDNA进行测序,建立系统发育树,进行系统学分析.结果从浙江舟山沿海海泥、海藻中分离到产淀粉酶的放线菌菌株43株,其中产淀粉酶能力较高的3株.放线菌A24产淀粉酶能力经选育后有较大提高:从初始酶活力111.5 U • mL-1经紫外诱变增至282.7 ...
