干扰素在体外对放疗后鼻咽癌病人自然杀伤细胞功能的影响 [PDF]
自然杀伤(NK)细胞在体内可以抑制肿瘤的发生,发展和转移,是机体执行免疫监视功能的重要效应细胞。NK细胞对肿瘤细胞的杀伤作用受到很多因素影响和调节。最近Brovall等人报告,电离辐射在体外可以抑制人NK细胞的活性。为了解按受放疗的肿瘤病人NK细胞功能的改变,并寻找对这种变化进行调整的办法,我们进行了干扰素对放疗后鼻咽癌病人NK细胞功能影响的初步观察。

采用单个细胞水平细胞毒方法研究比较了正常人外周血自然杀伤(NK)细胞对不同靶细胞结合与杀伤活性的差别。发现NK细胞对人胚胎成纤维细胞和低分化鼻咽癌(CNE-2)细胞的结合率明显高于对人成骨肉瘤细胞(OS-732)和PLC/PRF/5人肝癌细胞,结合并杀伤新鲜切除人肝癌细胞,而对同一病人的癌周肝细胞则无作用;在同一株CNE-2细胞中,NK多选择性地与核浆比例大的靶细胞结合。以上结果说明NK细胞在发挥自然细胞毒过程中具有一定的选择性,选择性地结合并杀伤分化差或幼稚的靶细胞。

自然杀伤(NK)细胞是T细胞后具有临床应用前景的另一类抗肿瘤免疫细胞。NK 细胞的活性主要受其表面受体和免疫微环境的调控,胶质瘤较强的免疫抑制性肿瘤微环境使NK细胞免疫治疗效率较低。本综述从胶质瘤-NK细胞交互作用角度讨论NK细胞在脑胶质瘤免疫治疗中作用的最新研究进展,总结靶向NK 细胞的化合物、单克隆抗体和细胞因子疗法,重点讨论了基因修饰的NK细胞在胶质瘤免疫治疗的现状及趋势,以及胶质瘤细胞的相关免疫逃逸分子机制,为基于NK细胞的免疫治疗神经胶质瘤提供理论依据和新的思路。

肿瘤是威胁全球人类身体健康的一类重大疾病,其病因复杂,并且每年的发病人数众多[1]。白细胞介素18(IL-18),以前称为干扰素γ(IFN-γ)诱导因子,于IL-1家族,能够诱导IFN-γ产生、激活自然杀伤(NK)细胞和毒性T淋巴细胞(CTL)、诱导辅助T细胞(Th),从而在机体抗感染、抗炎以及抗肿瘤的过程中发挥重要作用。鉴于IL-18在各种肿瘤中均有不同程度的表达,检测其在血清或者组织中的表达水平可能对肿瘤的实验诊断具有参考价值。

Study of Pelo deficiency mediated antiviral activity [PDF]
果蝇是研究抗病毒天然免疫以及病毒与宿主相互作用的理想模型。我们以果蝇C病毒(DrosophilaCVirus,DCV)为病原体对已有的突变体果蝇库进行筛选,找到了一个对DCV感染更为抵抗的果蝇株并鉴定出其体内Pelo基因的表达量下降。通过基因敲除和表型回复实验,我们证实了Pelo基因的缺失的确可以使果蝇对DCV的感染更为抵抗。此外,DCV以及其他多种病毒的复制在Pelo基因敲除型果蝇中都变慢,说明Pelo基因缺陷可能介导了广谱的抗病毒免疫。 我们研究发现,Pelo并不参与目前已知的果蝇抗病毒免疫途径 ...
