靶向抑制生长分化因子3减轻内皮细胞炎症反应并抑制脉络膜新生血管
目的揭示靶向抑制生长分化因子3(GDF3)对脉络膜新生血管(CNV)形成及内皮炎症反应的影响。方法构建激光诱导的小鼠CNV模型,通过RNA测序(RNA-seq)筛选差异基因。采用实时荧光定量PCR(RT-qPCR)和Western blot验证GDF3表达水平。通过siRNA及中和抗体拮抗内皮细胞GDF3,采用CCK8实验、划痕实验及成管实验评估内皮细胞增殖、迁移和血管生成能力。检测炎症/黏附分子表达变化,分析免疫细胞黏附和跨内皮迁移实验变化。在CNV模型中,通过玻璃体腔注射shRNA靶向抑制GDF3 ...
