高通量测序技术在慢性咽炎患者咽部细菌耐药基因检测中的应用价值

目的探讨高通量测序技术在慢性咽炎患者耐药基因分布中的应用。方法选取2010年5月至2012年5月本院门诊收治的慢性咽炎患者360例,采集患者咽后壁分泌物,提取质粒DNA并进行Solexa高通量测序,通过比对在NCBI质粒基因组数据库进行物种鉴定,通过与ARDB数据库进行比对进行耐药基因鉴定。结果本研究共鉴定出307个质粒,分属25种病原菌,所占比例最高的为α-溶血性链球菌(12.87%),其次为副流感嗜血菌(9.86%)。通过与ARDB中已知的380种ARG比对,共有4015条序列与已知耐药基因序列一致。

医学外显子组测序检测遗传病拷贝数变异的初步探索

【目的】评价医学外显子组高通量测序技术用于拷贝数变异(CNV)检测的可行性。【方法】两个不相关的遗传病家系案例,患者除了有智力障碍的类似症状外,无其他明显共同点。通过脆性X染色体综合征基因筛查、染色体G显带技术、染色体微阵列技术以及医学外显子组测序技术等对患者及其家属进行遗传学检测。【结果】在两个家系的患者中均发现染色体拷贝数变异,片段大小从11.4kb到13.03Mb不等。拷贝数变异得到了其他技术的验证。【结论】医学外显子组高通量测序技术用于拷贝数变异相关的遗传病临床辅助诊断是可行的 ...
