Interleukin (IL)-12 was originally identified as a factor produced by human Epstein-Barr virus-transformed B cell lines. It was detected by one group as cytotoxic lymphocyte maturation factor, a cytokine that synergized with IL-2 in the induction of lymphokine-activated killer cells and cytotoxic T lymphocytes.

Abstract cDNAs for human and murine IL-12 subunits have been cloned (Wolf et al. 1991; Gubler et al. 1991; Schoenhaut et al. 1992). The nucleotide sequences of the p40 subunit from man and mouse (GenBank accession numbers M65292, M65272, and M86671) predict polypeptides of 328 and 335 amino acids, respectively.

Abstract Interleukin-12 (IL-12) is a newly characterized cytokine that has a unique heterodimeric structure. It was initially cloned from B lymphoblastoid cell lines, but the majority of IL-12 is produced by macrophages/monocytes following appropriate stimulation. IL-12 can (1) enhance the cytolytic activity of a number of effector cells
