SPHINX31 acts as a SRPK1 inhibitor targeting the ATR/DNA‐PKcs/CHK1 replicative checkpoint to inhibit cell growth in non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
The kinase SRPK1 directly interacts with the protein TOPBP1 and regulates the pre‐mRNA splicing of WIZ thereby contributing to the activation of the ATR/CHK1 replicative checkpoint in response to replicative stress. This allows cancer cells' genomic stability and survival.

Mutant p53R273H disrupts PDPK1 homodimerization and contributes to PDPK1 activation

open access: yesMolecular Oncology, EarlyView.
How mutant p53R273H drives AKT signaling is unclear. We show that p53R273H, but not wild‐type, directly binds PDPK1 via a mutation‐dependent conformational change. This interaction disrupts inhibitory PDPK1 homodimerization and enhances AKT phosphorylation.

Peculiar Velocity Reconstruction from Simulations and Observations Using Deep Learning Algorithms

open access: yesThe Astrophysical Journal
In this paper, we introduce a U-Net model of deep learning algorithms for reconstructions of the 3D peculiar velocity field, which simplifies the reconstruction process with enhanced precision.

Everted U-Net for 3D scene reconstruction and segmentation

open access: yesITM Web of Conferences
Abstract. The field of data science related to the processing of three-dimensional objects is becoming more and more relevant. After the successes in image processing, the apotheosis of which was the development of generative neural net-works, the intensification of efforts in the direction of three-dimensional data processing looks logical.

Characterization of therapeutic antibodies targeting lymphatic and vascular resistance factor for the treatment of hypervascularized and highly metastatic cancers

open access: yesMolecular Oncology, EarlyView.
LVRF (lymphatic and vascular resistance factor) is a VEGFA splice variant upregulated in response to anti‐angiogenic therapy that fosters tumor adaptation through enhanced vascularization, lymphangiogenesis, and metastatic dissemination. Targeting LVRF with fully human neutralizing antibodies disrupts these processes and limits resistant tumor growth ...

Reprogramming tumor cells with LCOR mRNA nanotherapy to enhance immunotherapy

open access: yesMolecular Oncology, EarlyView.
LCOR (Ligand‐dependent corepressor) mRNA encapsulated in polymeric nanoparticles restores LCOR activity in tumor cells, constraining aggressive tumor cell states and increasing tumor immunogenicity. This reprogramming makes tumor cells more vulnerable to T‐cell attack and enhances their response to immune‐checkpoint immunotherapy.

Tumour‐proximal deep serum proteomics identifies biomarkers associated with ovarian cancer

open access: yesMolecular Oncology, EarlyView.
New biomarkers are needed for early ovarian cancer detection. Ovarian (tumour‐proximal) and peripheral vein serum from healthy women and patients with benign or malignant tumours underwent nanoparticle‐enriched deep proteomics. Ovarian serum yielded more protein identifications and revealed tumour‐reflective signatures.

Generalized U-Net for automatic liver segmentation and R2* estimation for assessment of iron overload using MRI

open access: yesBMC Medical Imaging
Background Although R2*-MRI is extensively validated for assessing liver iron concentration (LIC), techniques for robust and automatic liver extraction remain underdeveloped, limiting clinical workflow and adoption for automated LIC reporting. Hence this
