Dual PI3K/AKT and CDK4/6 inhibition reveals selective sensitivity in an SHH medulloblastoma stem cell model

open access: yesMolecular Oncology, EarlyView.
Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

Incremental analysis of constraint logic programs

open access: yes, 2000
Global analyzers traditionally read and analyze the entire program at once, in a nonincremental way. However, there are many situations which are not well suited to this simple model and which instead require reanalysis of certain parts of a program ...

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Correctness kernels of abstract interpretations

open access: yesInformation and Computation, 2014
In abstract interpretation-based static analysis, approximation is encoded by abstract domains. They provide systematic guidelines for designing abstract semantic functions that approximate some concrete system behaviors under analysis. It may happen that an abstract domain contains redundant information for the specific purpose of approximating a ...

Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Comparing Tag Scheme Variations Using an Abstract Machine Generator

open access: yes, 2008
In this paper we study, in the context of a WAM-based abstract machine for Prolog, how variations in the encoding of type information in tagged words and in their associated basic operations impact performance and memory usage.

A Quantum Formalism for Abstract Dynamical Systems

open access: yesMathematics
This paper presents a quantum formulation for classical abstract dynamical systems (ADS), defined by coupled sets of first-order differential equations. They are referred to as “abstract” because their dynamical variables can be of different interrelated

ESR1 methylation and ESR1 mutations in circulating tumor cells (CTCs) and paired plasma‐cfDNA of advanced breast cancer patients: A feasibility proof‐of‐concept study

open access: yesMolecular Oncology, EarlyView.
Circulating tumor cells (CTCs) and plasma cell‐free DNA (cfDNA) were analyzed to detect ESR1 mutations and methylation in patients with advanced breast cancer. CTC‐derived DNA showed higher sensitivity for mutation detection and revealed complementary genetic and epigenetic alterations, highlighting the added value of CTC analysis for understanding ...
