EGFR: New Insights on Its Activation and Mutation in Tumor and Tumor Immunotherapy

This review provides new perspectives on EGFR's activation, mutations, and their impact on tumor immunotherapy. It summarizes how EGFR is activated and the current EGFR targeting strategy, evaluates their clinical outcomes in combination with immunotherapy, highlighting differential responses in tumors with EGFR wild‐type and different mutants.

APOL2 Stabilizes Ku80 to Confer NHEJ‐Mediated Radioresistance in Gastric Cancer

Radiotherapy resistance in gastric cancer (GC) is often mediated by enhanced DNA repair. APOL2 is found to promote NHEJ by stabilizing Ku80, conferring radioresistance. High APOL2 correlates with poor prognosis. Formononetin disrupts APOL2‐Ku80, restoring radiosensitivity, suggesting a novel therapeutic strategy.

An MRI‐derived Lymph Nodes morphological and topological structure (LNs‐MTS) model is established to evaluate the immune status of the lymph node system in rectal cancer. It provides a non‐invasive, quantitative approach for prognosis assessment and optimizing adjuvant therapy decisions.

Exosomal CCT6A Secreted by Cancer‐Associated Fibroblasts Interacts with β‐Catenin to Enhance Chemoresistance and Tumorigenesis in Gastric Cancer

Here, it is revealed that CAF‐secreted exosomal CCT6A enhances stemness, chemoresistance, and glycolysis in GC via the β‐catenin/c‐Myc/DDIT4‐TXNIP axis. Furthermore, CCT6P1, a pseudogene of CCT6A, is identified as upregulating CCT6A by competitively binding miR‐922, while c‐Myc transcriptionally activates both CCT6P1 and CCT6A.

Developing a Personalized Cancer Nanovaccine Using Coxsackievirus‐Reprogrammed Cancer Cell Membranes for Enhanced Anti‐Tumor Immunity

A personalized nanovaccine combining PLGA‐encapsulated heat‐inactivated CVB3 and membranes from CVB3‐infected breast cancer cells stimulates potent antitumor immunity. It reduces immunosuppressive markers, enhances immune activation, and improves survival in vivo.

Patients‐Derived Organoids Sequencing‐based FOXP4 Facilitates Radioresistance by Transcriptionally Modifying GPX4 to Regulate ferroptosis in Colorectal Cancer

FOXP4 regulates GPX4 transcription to suppress ferroptosis. In CRC patients, initial biopsies established PDO cultures alongside concurrent chemoradiotherapy. Post‐treatment MRI with mrTRG scoring stratified radioresistant/sensitive cohorts. Parallelly, 8Gy‐irradiated PDOs underwent viability‐based stratification.
