Elucidation of interface interactions between a dehydratase domain and an acyl carrier protein in cremimycin polyketide synthase

open access: yesFEBS Letters, EarlyView.
In modular polyketide synthases, the dehydratase (DH) domain catalyzes the dehydration reaction of the β‐hydroxyacyl unit attached to the cognate acyl carrier protein (ACP) domain. However, it is unclear how DH interacts with ACP during the reaction. In this study, we identified DH–ACP interface residues, providing the first detailed insights into DH ...

Functional variation among LPMOs revealed by the inhibitory effects of cyanide and buffer ions

open access: yesFEBS Letters, EarlyView.
This study addresses the inhibition of lytic polysaccharide monooxygenases (LPMOs) by cyanide and explains how and why the magnitude of observed inhibitory effects depends on the way LPMO reactions are setup and on the type of LPMO. Enzymes known as lytic polysaccharide monooxygenases (LPMOs) are mono‐copper polysaccharide‐degrading peroxygenases that ...

Measuring clinical outcomes in adult ADHD clinics: psychometrics of a new scale, the adult ADHD Clinical Outcome Scale

open access: yesBJPsych Open
Background Adult attention-deficit hyperactivity disorder (ADHD) clinics are in their infancy in Ireland and internationally. There is an urgent need for clinical evaluation of these services. Until now, clinical outcomes have relied mainly on functional

Ablation of LRP6 in alpha‐smooth muscle actin‐expressing cells abrogates lung inflammation and fibrosis upon bleomycin‐induced lung injury

open access: yesFEBS Letters, EarlyView.
Low‐density lipoprotein receptor‐related protein 6 (LRP6) is a key receptor for the Wnt antagonist Dickkopf1 (DKK1). DKK1 protein expression is induced in a bleomycin (BLM)‐induced lung injury model. We show that DKK1 induces proinflammatory and profibrotic genes in lung fibroblasts.

A crossroad in ADHD – adult-onset ADHD

open access: yesEuropean Psychiatry, 2017
IntroductionAttention-deficit/hyperactivity disorder (ADHD) is a common disorder characterized by inattention or hyperactivity–impulsivity, or both. For a long time, ADHD was thought of as a disorder of children which would sometimes persist into adulthood.

Characteristics of the Kelch domain containing (KLHDC) subfamily and relationships with diseases

open access: yesFEBS Letters, EarlyView.
The Kelch protein superfamily includes 63 members, with the KLHDC subfamily having 10 proteins. While their functions are not fully understood, recent advances in KLHDC2's structure and role in protein degradation have highlighted its potential for drug development, especially in PROTAC therapies.

Finding essential parts of the brain in rs-fMRI can improve diagnosing ADHD by Deep Learning [PDF]

open access: yesarXiv, 2021
Attention Deficit\Hyperactivity Disorder(ADHD) is considered a very common psychiatric disorder, but it is difficult to establish an accurate diagnostic method for ADHD. Recently, with the development of computing resources and machine learning methods, studies have been conducted to classify ADHD using resting-state functional magnetic resonance ...

Insertion of the FeB cofactor in cNORs lacking metal inserting chaperones

open access: yesFEBS Letters, EarlyView.
Nitric oxide reductase is an enzyme found in the bacterial denitrification pathway. The NOR active site contains a non‐heme iron, often, but not always inserted with the assistance of chaperones. Here, we study the insertion of FeB in the subfamily of cNORs lacking chaperones and found a putative channel, conserved in the family, perhaps enabling the ...

ADHD diagnosis based on action characteristics recorded in videos using machine learning [PDF]

open access: yes
Demand for ADHD diagnosis and treatment is increasing significantly and the existing services are unable to meet the demand in a timely manner. In this work, we introduce a novel action recognition method for ADHD diagnosis by identifying and analysing raw video recordings. Our main contributions include 1) designing and implementing a test focusing on
