酵素をその活性を保ったままで不溶化し,基質と反応させようとする考え方は,酵素の研究,利用という面から多くの利点を与えるものと考える。著者らはその方法の一つとして吸着剤に酵素を吸着させることによって,水に不溶なcomplexを調製しているが,本報では被吸着酵素の動態を考察するためにMichaelis定数および最大速度を測定した。すなわち,酵素としては市販の糖化型アミラーゼを使用し,これを活性炭または酸性白土に吸着させてcomplexを調製し,これを種々な濃度の基質と反応させたところ,Michaelis定数はいずれもnative酵素より増加し,最大速度は減少した。これにより,吸着剤表面の酵素はかなり構造上の変化を受けていることがわかった。また吸着剤の種類によって ...
