Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Matrix metalloproteinase‐9 regulates cell adhesion and membrane protrusive activity of ovarian cancer cells

open access: yesFEBS Open Bio, EarlyView.
Matrix metalloproteinase‐9 (MMP9) drives ovarian cancer progression. Using MMP9‐null cells (M9‐KO) created from ovarian cancer cells, we found MMP9 loss did not block Epidermal Growth Factor (EGF)‐driven E‐cadherin dissolution or EMT but delayed and reduced EGF‐driven membrane protrusions. Transient MMP9 re‐expression drove membrane protrusion.

Abundance and distribution of narwhals (Monodon monoceros) on the summering grounds in Greenland between 2007-2019

open access: yesFrontiers in Marine Science
Narwhal abundance in West Greenland (WG) and East Greenland (EG) was estimated from aerial surveys conducted between 2007 and 2019 at their summer grounds.

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Drones, sensors, and their potential utility for mountain sheep surveys

open access: yesCalifornia Fish and Wildlife Journal
Unmanned aerial vehicles (UAV), often referred to as unmanned aerial systems (UAS) or drones, are experiencing increasing use by wildlife researchers and for a variety of purposes. When equipped with one or more of the various sensors currently available,

Abundance and density of lesser prairie‐chickens and leks in Texas

open access: yesWildlife Society Bulletin, 2013
Lesser prairie‐chickens (LEPCs; Tympanuchus pallidicinctus) have experienced population declines due to both direct and indirect habitat loss, including conversion of native rangeland to cropland and disturbance from energy development.

Mass spectrometry based identification of AMP‐O‐Tris generated by Thermococcus onnurineus Cas10

open access: yesFEBS Open Bio, EarlyView.
Isolated Thermococcus onnurineus Cas10 generates the noncanonical ATP‐derived product AMP‐O‐Tris while in Tris‐containing buffer as identified via mass spectrometry, revealing relaxed nucleophile selectivity under isolated conditions. These findings suggest that multiprotein Csm complex assembly restricts Cas10 reactivity toward canonical cyclic ...

Sea turtle nesting in the Ten Thousand Islands of Florida [PDF]

open access: yes, 1997
Loggerhead sea turtles (Caretta caretta) nest in numerous substrate and beach types within the Ten Thousand Islands (TTl) of southwest Florida. Nesting beach selection was analyzed on 12 islands within this archipelago.

Whole‐Body Pattern of Muscle Degeneration and Progression in Sarcoglycanopathies

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective To characterize whole‐body intramuscular fat distribution pattern in patients with sarcoglycanopathies and explore correlations with disease severity, duration and age at onset. Methods Retrospective, cross‐sectional, multicentric study enrolling patients with variants in one of the four sarcoglycan genes who underwent whole‐body ...
