Abstract Acute leukemia with t(4;11)(q21,q23) translocation results from the in-frame fusion of the MLL to the AF4/FEL gene. In previous studies, we and others demonstrated that AF4 transcripts are present in a variety of hematopoietic and nonhematopoietic human cells.

Southern anal. of the MLL Locus in DNA from bone marrow cells samples from a 5-mo old child with acute lymphoblastic leukemiasampled at diagnosis revealed a rearrangement at locus MLL of chromosome 11q23. Panhandle PCR amplification of the fragment comprising the der(11) translocation breakpoints yielded a 4.6 kb fragment corresponding to the bamHI ...

Alert and sniffy-nosed about Mll-AF4?
Blood, 2006Comment on Chen et al, page [669][1] A report from the Kersey laboratory documents the long-awaited successful establishment of a murine model for leukemogenesis by the Mll-AF4 fusion gene. The MLL gene is notorious for its promiscuous fusion with multiple alternative partner genes in leukemia,

The ongoing conundrum of MLL-AF4 driven leukemogenesis
Blood, 2013In this issue of Blood, Bueno and colleagues explore the developmental impact, as well as the transforming capacity, of the mixed-lineage leukemia (MLL)–AF4 fusion protein in combination with activation of FMS-like tyrosine receptor 3 (FLT3) in human embryonic stem cells (hESCs ...

MLL-AF4 Gene Fusions in Normal Newborns
Blood, 1999To the Editor: Uckun et al[1][1] have reported that bona fide in-frame fusion MLL-AF4 fusion sequences are detectable, using a sensitive nested reverse transcription-polymerase chain reaction (RT-PCR) method, in around 25% of normal fetal bone marrow and liver samples and in 1 of 6 infant ...

Differential expression of AF4/FEL mRNA in human tissues
Genetic Analysis: Biomolecular Engineering, 1996This manuscript reports the differential expression of the AF4 gene among human tissues. AF4 mRNA is highly expressed in normal placental tissue which correlates with the newborn age of patients presenting with leukemia characterized by the MLL/AF4 gene rearrangement.

Detection of MLL/AF4 Recombination by PCR Technique
1996The molecular analysis of chromosomal translocations occurring in human leukemias revealed the existence of newly formed fusion genes. The balanced translocation between chromosomes 4 and 11, for instance, results in a new MLL/AF4 gene as the AF4 gene at chromosomal region 4q21 is juxtaposed next to the part of the MLL gene (also called ALL-1 or HRX ...
