Oligomeric States and Hydrodynamic Properties of Lysyl Oxidase-Like 2

open access: yesBiomolecules, 2021
Lysyl oxidase-like 2 (LOXL2) has emerged as a promising therapeutic target against metastatic/invasive tumors and organ and tissue fibrosis. LOXL2 catalyzes the oxidative deamination of lysine and hydroxylysine residues in extracellular matrix (ECM ...

Simple Determination of Gold Nanocrystal Dimensions by Analytical Ultracentrifugation via Surface Ligand-Solvent Density Matching

open access: yesNanomaterials, 2021
Analytical ultracentrifugation (AUC) is a powerful technique to observe colloidal nanocrystals (NCs) directly in solution and obtain critical information about their physical-chemical properties.

A coiled‐coil domain triggers oligomerization of MmpL10, the mycobacterial transporter of trehalose polyphleate precursor

open access: yesFEBS Letters, EarlyView.
Most MmpLs transport lipids and glycolipids of the complex mycomembrane of mycobacteria. This study shows that MmpL10 from Mycobacterium smegmatis, the transporter of the trehalose polyphosphate precursor, harbors a coiled‐coil‐like extension. Biochemical and electron microscopy studies demonstrate that the coiled‐coil enables MmpL10 to trimerize.

Formation of Nanoclusters in Gold Nucleation

open access: yesCrystals, 2020
Gold nanoclusters consisting of a specific atom number have gained popularity in research in recent years due to their outstanding properties. Due to their molecule-like behavior, their properties depend strongly on their size.

Structural Characterization of the Extracellular Domain of CASPR2 and Insights into Its Association with the Novel Ligand Contactin1 [PDF]

open access: yes, 2015
Contactin-associated protein-like 2 (CNTNAP2) encodes for CASPR2, a multidomain single transmembrane protein belonging to the neurexin superfamily that has been implicated in a broad range of human phenotypes including autism and language impairment.

Thermal proteome profiling and proteome analysis using high‐definition mass spectrometry demonstrate modulation of cholesterol biosynthesis by next‐generation galeterone analog VNPP433‐3β in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Elevated level of cholesterol is positively correlated to prostate cancer development and disease severity. Cholesterol‐lowering drugs, such as statins, are demonstrated to inhibit prostate cancer. VNPP433‐3β interrupts multiple signaling and metabolic pathways, including cholesterol biosynthesis, AR‐mediated transcription of several oncogenes, mRNA 5′

pH Dependence and Stoichiometry of Binding to the Fc Region of IgG by the Herpes Simplex Virus Fc Receptor gE-gI [PDF]

open access: yes, 2004
Herpes simplex virus type 1 encodes two glycoproteins, gE and gI, that form a heterodimer on the surface of virions and infected cells. The gE-gI heterodimer has been implicated in cell-to-cell spread of virus and is a receptor for the Fc fragment of IgG.

Resuspending samples in analytical ultracentrifugation

open access: yesAnalytical Biochemistry, 2020
In analytical ultracentrifugation it is often very useful to resuspend samples in situ after sedimentation experiments for further investigation. This can be achieved by manually subjecting the entire sample cell assembly to gentle motion that causes the air bubble in the sample compartment to repeatedly move through the solution and thereby cause ...

CNS Mitochondria‐Derived Vesicle in Blood: Potential Biomarkers for Brain Mitochondria Dysfunction

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Mitochondrial dysfunction is a hallmark of neurodegenerative diseases like Alzheimer's (AD) and Parkinson's (PD). Our goal was to develop practical, noninvasive methods to assess mitochondrial status through the detection of mitochondria‐derived vesicles (MDVs).

Structure and Activation Mechanism of the Drosophila Initiator Caspase Dronc [PDF]

open access: yes, 2006
Activation of an initiator caspase is essential to the execution of apoptosis. The molecular mechanisms by which initiator caspases are activated remain poorly understood.
