Dual PI3K/AKT and CDK4/6 inhibition reveals selective sensitivity in an SHH medulloblastoma stem cell model

open access: yesMolecular Oncology, EarlyView.
Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

Approximate Computing with Probabilistic Programs [PDF]

open access: yes, 2015
Approximate computing involves relaxing program accuracy requirements to improve performance or decrease energy consumption. Since program accuracy measures tend to be non-deterministic due to multiple sources of uncertainty (e.g., inputs), it should be

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

VLSI Circuits for Approximate Computing [PDF]

open access: yes, 2017
Approximate Computing has recently emerged as a promising solution to enhance circuits performance by relaxing the requisite on exact calculations. Multimedia and Machine Learning constitute a typical example of error resilient, albeit compute-intensive,

Design and Implementation of Approximate Hybrid Floating-Point Multipliers for Signal-Processing Applications

open access: yesIEEE Access
Floating-point (FP) multipliers are critical components in high-performance computing systems, particularly in signal processing, graphics, and machine learning applications. However, traditional FP multipliers face challenges in power, area, and latency

Approximate Computing for Test and Test of Approximate Computing

open access: yes
Approximate Computing (AxC) is nowadays a well established design and computing paradigm to produce more efficient computation systems by judiciously reducing the computation quality. In particular, AxC has been successfully applied to Integrated Circuits (ICs), in the last years in both ways: AxC for test and the test of AxC integrated circuits.

Hippo pathway at the crossroads of stemness and therapeutic resistance in breast cancer

open access: yesMolecular Oncology, EarlyView.
Dysregulation of the Hippo pathway drives nuclear accumulation of YAP/TAZ, activating stemness‐related transcriptional programs that sustain breast cancer stemness and fuel therapeutic resistance across subtypes, underscoring Hippo signaling as a targetable vulnerability. Figure created and edited with BioRender.com.

Deciphering transcriptional plasticity in pancreatic ductal adenocarcinoma reveals alterations in sensory neuron innervation

open access: yesMolecular Oncology, EarlyView.
Pancreatic sensory neurons innervating healthy and PDAC tissue were retrogradely labeled and profiled by single‐cell RNA sequencing. Tumor‐associated innervation showed a dominant neurofilament‐positive subtype, altered mitochondrial gene signatures, and reduced non‐peptidergic neurons.

Approximate computing of reliability characteristics of some reserved systems

open access: yesNauka ta progres transportu, 2012
In work two redundant systems with restored elements are investigated: with a warm sliding reserve, control and preventive maintenance, and a hot sliding reserve. The software for the approached calculation of some characteristics of reliability of these

Differential expression of cancer‐related genes supports prediction of poor response to first‐line treatments in T‐ALL pediatric patients with high minimal residual disease

open access: yesMolecular Oncology, EarlyView.
In the present work, we have identified a transcriptional signature based on the differential expression of six genes (BCL2&MAST4, HSH2D&LAT2, METRN&PITPNM2) that would facilitate the early detection of T‐cell acute lymphoblastic leukemia (T‐ALL) patients prone to a poor treatment response and could be implemented at diagnosis, along with other risk ...
