Skin tape strip lipidomic analysis revealed preexisting skin barrier lipid abnormalities at day 0 that were further exacerbated after 6 weeks of CPI treatment. Significant changes in skin (but not plasma) proinflammatory cytokines were identified in future ircAE patients after 6 weeks of CPI immunotherapy.

Shotgun Metagenomics Reveals Skin Microbiome Composition and Function in Infant Atopic Disease
Deep shotgun metagenomics profiled > 1000 infant skin swabs in the VITALITY cohort. Distinct skin microbiome signatures separated AD alone from AD with allergic co‐occurrence. Early Staphylococcus enrichment preceded later AD with allergic co‐occurrence. FLG mutations and strain‐level analyses revealed host and microbial features linked to AD.

Upon activation, they rapidly release preformed granule‐associated mediators including histamine and lipid mediators such as LTC4, while cytokine production occurs over a longer timescale, contributing to downstream immune responses.

Dietary Habits and Atopic Dermatitis Significantly Influence the Fecal Microbiome
Atopic dermatitis, dietary habits, sex, and BMI significantly influenced the fecal microbiome composition. On the species level, Faecalibacterium taiwanense was more abundant in atopic dermatitis, and Vescimonas sp000435555 was more abundant in the control population. On the genus level, CAG‐302 (NCBI: Clostridium) was more abundant in controls.
