AbstractFormylglycine‐generating enzymes provide a convenient tool for site‐specific protein derivatization. Their ability to oxidize cysteine or serine residues within a defined consensus sequence to Cα‐formylglycine (FGly) allows for the targeted introduction of a unique chemical handle for various bioconjugation reactions.
