In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Influence of the physical properties of the liquid on axial dispersion in packed columns

open access: yes
The effects of the viscosity and surface tension on the axial dispersion and liq. holdup was investigated in a 0.3-m-diam. packed column with countercurrent gas-liq. flow.

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Reduced-Order Modeling of Indirect Fluidized-Bed Particle Receivers with Axial Dispersion

open access: yesSolarPACES Conference Proceedings
Oxide particles present a heat transfer and thermal energy storage (TES) media for next-generation concentrating solar power (CSP) plants where the high-temperature particle TES can provide dispatchable solar power [1].

Axial dispersion of the liquid phase in vertical tubular contactors with static mixers

open access: yes, 1986
Axial dispersion characteristics in the liquid phase have been determined for three selected types of static mixers using the impulse response technique in a vertical tubular contactor for single phase (water) and two phase (water-air, cocurrent flow ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

YlmG1 is localized exclusively to the chloroplast envelope membrane and is involved in preprotein translocation in Arabidopsis thaliana

open access: yesFEBS Open Bio, EarlyView.
Cytosolically synthesized chloroplast preproteins are translocated across the outer and inner envelope membranes through translocons called TOC and TIC, respectively. In green algae and plants, the TIC core is composed of essential membrane proteins, Tic12, Tic20, and Tic214.

Estimating intra-axonal axial diffusivity with diffusion MRI in the presence of fibre orientation dispersion

open access: yes, 2020
Intra-axonal axial diffusivity could be interesting biomarker of disease, yet it is often assumed constant across the white matter. Furthermore, when intra-axonal diffusivity is estimated, few models account for fibre orientation dispersion which (when ...

Identifying transcription factors controlling the basal expression of human MRP4 highlights a substantial role for Sp1

open access: yesFEBS Open Bio, EarlyView.
The MRP4 transporter exports several drugs and signaling molecules. Here, we identified key promoter elements regulating basal MRP4 expression. Using reporter assays, we defined a conserved region with essential Sp1 and contributory Ets sites, which controlled basal MRP4 expression.
