Evolution‐guided yeast complementation reveals functional differences in human PSPH variants

open access: yesFEBS Open Bio, EarlyView.
Ancient genomes can help guide which human genetic variants are tested experimentally. This study applies that idea to PSPH, a gene involved in serine biosynthesis, and uses high‐throughput yeast complementation to compare variant function. The findings reveal measurable differences among selected alleles and illustrate the value of evolution‐guided ...

Behavioral inhibition and cognitive control in an emotional Go/No-go task

open access: yes
Behavioral inhibition is a temperamental tendency associated with heightened risk for developing anxiety disorders. Individuals with behaviorally inhibited temperament tend to avoid or withdraw from novel situations, which may be associated with ...

In silico and in vitro exploration of a tyrosinase for biocatalytic production of catechols

open access: yesFEBS Open Bio, EarlyView.
Tyrosinase from Ralstonia pseudosolanacearum is a promising biocatalyst for producing valuable catechols from monophenol substrates. This tyrosinase is uniquely suited to this due to its high monophenolase : diphenolase ratio. We combined in silico docking and in vivo kinetic characterisation of this tyrosinase with 11 industrially relevant monophenols,

Purification and preparation of Marchantia polymorpha Auxin Response Factor 2 for phase separation studies

open access: yesFEBS Open Bio, EarlyView.
We describe detailed protocols for the purification and preparation of Marchantia polymorpha Auxin Response Factor 2 (MpARF2). This protein is fused to an MBP solubility tag and an mNG fluorescent tag and is purified from Escherichia coli. The presented procedures make it possible to study MpARF2 assemblies, which could arise from phase separation ...

Heterotropic regulation and negative homotropic cooperativity

open access: yesFEBS Open Bio, EarlyView.
We identified a structural module common to some proteins that couple negative cooperativity with heterotropic regulation, two features that rarely coexist. These proteins are ring‐like and present an ordered asymmetry whereby noncontacting subunits are symmetric, and their tertiary structure differs from that of contacting subunits.

Behavioral Inhibition as a Possible Etiology of the Anxiety Disorders

open access: yes, 1996
iv, 42 p.Behavioral Inhibition is a syndrome that is characterized by shyness, and withdrawal from an unfamiliar person or situation. It has behavioral and physiological components that can be identified in 21 month old children and remain fairly ...

The C‐terminal truncated splicing variant of NK1R negatively modulates substance P‐stimulated NK1R signaling

open access: yesFEBS Open Bio, EarlyView.
The neurokinin 1 receptor exists as full‐length (NK1L) and C‐terminally truncated (NK1S) splice variants. We show that NK1S heterodimerizes with NK1L, impairing Gαq coupling and Ca2+ mobilization while enhancing β‐arrestin1 recruitment. NK1S suppresses substance P‐driven gene expression and cell migration, revealing NK1S as an endogenous biased ...

Predicting Generalized Anxiety Disorder Based on Emotion Regulation Deficits, Thought-Action Fusion, and Behavioral Inhibition [PDF]

open access: yes
Background & Aims: Generalized anxiety disorder (GAD) can be affected by different emotional, cognitive, and natural factors. The purpose of this study was to predict GAD based on emotion regulation deficits, thought-action fusion, and behavioral ...

Translational Approaches to the Neurobiological Study of Conditional Discrimination and Inhibition: Implications for Psychiatric Disease

open access: yes
There is a growing number of studies investigating discriminatory fear conditioning and conditioned inhibition of fear to assess safety learning, in addition to extinction of cued fear.

Type I interferons modulate autophagy to shape gemcitabine response in pancreatic cancer cells

open access: yesFEBS Open Bio, EarlyView.
Type I interferons differentially modulate autophagy and the response of pancreatic cancer cells to gemcitabine. IFNα2b stimulates autophagic flux and protects cells from gemcitabine‐induced cell death, contributing to chemoresistance. In contrast, IFNβ1a inhibits autophagosome formation and enhances gemcitabine‐induced cell death, resulting in ...
