Site‐specific biotinylation

International Journal of Peptide and Protein Research, 1992
We have developed an expeditious method for the incorporation of the biotinylaminocaproyl moiety on the ε‐amino group of a lysine residue within a peptide chain in a site‐specific manner. Using t‐Boc chemistry for the solid phase synthesis approach and a base labile, acid stable protecting group (Fmoc‐) for the ε‐amino group of the target lysine, we ...

[14] Protein biotinylation

1990
Publisher Summary This chapter discusses the procedures by which researchers have incorporated biotin moieties into proteins. Several laboratories have reported the use of an avidin-based assay for detecting the extent of biotinylation. An effective method for detecting the average number of biotin groups per protein molecule would be to first remove

Biotinylated peptides/proteins. II. Identification of biotinylated lysyl phenylthiohydantoins.

Analytical biochemistry, 1991
The identification and characterization of biotinylated lysyl residues in a polypeptide chain by automated sequence analysis is described. An in depth analytical study was conducted for the delineation of N epsilon modification of lysyl residues with N-hydroxysuccinimide esters of biotin and 6-aminohexanoic biotin.

Selections on Biotinylated Antigens

2001
Phage antibody library selections on peptides or proteins are usually carried out using antigens directly coated on a plastic surface (e.g. Petri dishes, microtiter plate well, immunotubes, Chapter 9). This straightforward method is easy to perform and has been shown to be very successful for a diverse set of antigens (for review see Winter et al. 1994)

Kinase-Catalyzed Biotinylation.

Current protocols in chemical biology
Kinase-catalyzed protein phosphorylation plays an essential role in a variety of biological processes. Methods to detect phosphoproteins and phosphopeptides in cellular mixtures will aid in cell biological and signaling research. Our laboratory recently discovered the utility of γ-modified ATP analogues as tools for studying phosphorylation ...
