CRISPR Enabled Precision Oncology: From Gene Editing to Tumor Microenvironment Remodeling

CRISPR technology has progressed from a prokaryotic immune system to a diverse suite of editing platforms, including Cas nucleases, base and prime editors, and RNA‐targeting enzymes. These advances enable precise genomic and epigenomic interventions, high‐throughput functional screening, and immune engineering.

Abstract Background Membrane contact sites are crucial for the exchange of ions or lipids and thus are critical for the function and maintenance of organelles. VPS13A is a membrane‐residing, bridge‐like protein connecting two membranes to enable bulk lipid transfer. Loss‐of‐function mutations in the VPS13A gene cause VPS13A disease.

In this study, we developed an interpretable machine learning (ML) ensemble framework and, by integrating the VEC criterion with the proposed machine learning scoring parameter in the alloy composition screening process, successfully designed multiple CoCrFeNiMn‐based HEAs with TWIP/TRIP effects and without the BCC phase.
