Assessing Caspase-1 Activation
2016The caspase-1 enzymatic activity plays a major role in the innate immune response as it regulates the maturation of two major proinflammatory cytokines, the interleukin-1beta (IL-1β) and IL-18. In this chapter, we describe the technique of Western blot to assess caspase-1 activation. This method provides multiple information within one experiment.

Immunoblotting for Active Caspase-1
2013Immunoblotting for caspase-1 is the gold-standard method of detecting inflammasome activation. In contrast to IL-1β-based readouts, it can be used in an experimental setup independent of de novo gene expression. Here, we present protocols for the preparation and precipitation of supernatant samples containing activated caspase-1 as well as protocols ...

The intricate biophysical puzzle of caspase-1 activation
Archives of Biochemistry and Biophysics, 2021This review takes a closer look at the structural components of the molecules involved in the processes leading to caspase-1 activation. Interleukins 1β and 18 (IL-1β, IL-18) are well-known proinflammatory cytokines that are produced following cleavage of their respective precursor proteins by the cysteine protease caspase-1.

Sennoside A is a novel inhibitor targeting caspase-1
Food & Function, 2022Sen A decreases caspase-1 activity and P2X7 function, which further leads to NLRP3 and AIM2 inflammasome disassembly and the reduced secretion of IL-1β and IL-18viapyroptosis restraining and NF-κB-involved NLRP3 priming.

Caspase-1: an integral regulator of innate immunity
Seminars in Immunopathology, 2015Caspase-1 is a unique cysteine protease playing central roles in innate immunity. Pathogens, stress, and damage signals induce activation of caspase-1, typically mediated by proximity-induced autoproteolysis in multimeric protein complexes called the inflammasome. Active caspase-1 induces secretion of pro-inflammatory cytokines and mediates pyroptosis,

Obesity development in caspase-1-deficient mice
International Journal of Obesity, 2013Caspase-1 is a member of the intracellular cysteine protease family that mediates inflammation through the activation of the cytokines interleukin-1β (IL-1β) and interleukin-18 (IL-18). As mice lacking IL-18 become obese and insulin resistant, and both IL-18 and IL-1β have a role in overall energy balance, we sought to determine whether caspase-1 ...

Nucleotide Sequence of Equine Caspase-1 cDNA
DNA Sequence, 1999Caspases are a family of cysteine proteases which have important roles in activation of cytokines and in apoptosis. Caspase-1, or interleukin-1 beta converting enzyme (ICE), promotes maturation of interleukin-1 beta (IL-1 beta) and interleukin-18 (IL-18) by proteolytic cleavage of precursor forms to generate biologically active peptides.

Quantifying Caspase-1 Activity in Murine Macrophages
2018The caspase-1 protease is a core component of multiprotein inflammasome complexes, which play a critical role in regulating the secretion of mature, bioactive pro-inflammatory cytokines interleukin (IL)-1β and IL-18. The activity of caspase-1 is often measured indirectly, by monitoring cleavage of cellular caspase-1 substrates, processing of caspase-1 ...

Sevoflurane is one of the most commonly used volatile anesthetics. Recent studies have shown that sevoflurane plays an important role in modulation of inflammation and immunity. However, little is known about the related molecular mechanisms. This study was designed to investigate the effects and mechanisms of sevoflurane on inflammatory cell death ...
