The cyclin-dependent kinases, most notable of which is cdc2, are key regulators of the cell cycle, and are highly conserved in evolution. We have cloned and analysed three cdc2-related kinase-encoding genes (tbcrk1-3) from the protozoan parasite Trypanosoma brucei.

Downregulation of the cdc2/Cyclin B Protein Kinase Activity by Binding of p53 to p34cdc2

We previously found that p53 binds to the catalytic subunit of the p34(cdc2)/cyclin B1-kinase. In the present study we analyzed the functional consequences of this interaction. Binding of wild-type p53 to p34(cdc2)/cyclin B1 results in a significant decrease of its histone H1 kinase activity.

A homolog of the fission yeast cdc2-encoded protein kinase (p34) is a component of M phase promoting factor in Xenopus oocytes. The homologous kinase in human HeLa cells is maximally active during mitosis, suggesting a mitotic role in mammalian somatic cells.
