A chitosanase from Paecilomyces lilacinus with binding affinity for specific chito‐oligosaccharides
Biotechnology and Applied Biochemistry, 2005A purple‐spore, rhizosphere‐inhabiting nematophagous fungus, further identified as Paecilomyces lilacinus, was found to grow on chitosanase‐detecting plate. An induced endochitosanase having a molecular mass of 23 kDa was purified from the culture medium by a single cation‐exchange column‐chromatography step.

We describe the synthesis of N-trifluoroacetyl chito-oligosaccharides and their use as ligands to probe the binding sites of wheat-germ agglutinin, a lectin specific for N-acetylglucosamine. The binding is monitored using intrinsic protein fluorescence, which is due to tryptophan side-chains.
