Determination of the structure of agarose gels by gel chromatography

Abstract Well-characterized fractions of the synthetic polysaccharide Ficoll have been chromatographed on columns of pearl-condensed agarose gels of concentrations between 2 and 8%. The results agree with the theory that the Ficoll is sterically excluded from the gel, which is made up of a random three-dimensional network of long fibers.

Abstract The chromatographic behavior of several glycogens on the agarose gels Sepharose 2B and Sepharose 4B has been investigated. These are shown to be convenient media for examination of the polydispersity and relative molecular weights of different glycogens.

In 1959 two Swedish researchers Jerker Porath and Per Gustaf Flodin reported their findings in Nature in a paper entitled “Gel Filtration: a method for desalting and group separation” (1). They described a method based on a column procedure which they claimed was “similar” to chromatography.
