Enrichment of Full AAV2 Using Multicolumn Countercurrent Solvent Gradient Purification (MCSGP)

open access: yesBiotechnology and Bioengineering, EarlyView.
The multicolumn countercurrent solvent gradient purification (MCSGP) method was evaluated as a method to enhance the separation of empty and full capsids during AAV2 purification. The results demonstrated an enrichment of full AAV2 particles using MCSGP.

N‐ to C‐Glycoside Rearrangement of Uridine 5′‐Phosphate in Two Enzymatic Steps for the Production of Pseudouridine 5′‐Phosphate

open access: yesBiotechnology and Bioengineering, EarlyView.
The authors present a streamlined cascade reaction for one‐pot N‐C rearrangement of uridine‐5′‐phosphate (UMP) to pseudouridine‐5′‐phosphate (ΨMP; a key mRNA vaccine building block), in two enzymatic steps catalyzed by UMP nucleosidase (EC 3.2.2.10) and ΨMP C‐glycosidase (EC 4.2.1.70).

Integration of Single Pass Tangential Flow Filtration and High Performance Countercurrent Membrane Purification for Intensification of Monoclonal Antibody Processing

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT There is increasing interest in the development of intensified processes that can provide significant reductions in manufacturing costs for monoclonal antibody (mAb) products. This study examines the performance of an integrated membrane process that combines single‐pass tangential flow filtration (SPTFF) with high‐performance countercurrent ...

3D Culture in Functionalized FN‐Silk Networks Facilitate Proliferation, Differentiation and Phenotypic Stability of Mature Human Primary Cells and Stem Cells

open access: yesBiotechnology and Bioengineering, EarlyView.
The authors present a new technique for 3D culture of mammalian cells with the support of a microfiber‐based network of fibronectin (FN)‐functionalized recombinant silk. This feasible 2‐step method results in highly viable free‐floating 3D cultures with tissue‐mimicking morphology.

The P134Q Sucrose Phosphorylase: Subtle Changes in the Catalytic Properties Benefit the Production of 2‐O‐α‐Glucosyl Glycerol

open access: yesBiotechnology and Bioengineering, EarlyView.
Production of the cosmetic ingredient 2‐α‐d‐glucosyl glycerol (2GG) by the Pro134Gln variant of sucrose phosphorylase from Bifidobacterium adolescentis is reported. The authors show with detailed characterization and hybrid kinetic modeling that the enzyme variant can improve the 2GG productivity of the biocatalytic process due to its increased ...
