Angewandte Chemie International Edition, EarlyView.Pseudouridine‐5′‐triphosphate (ΨTP) and its N1‐methylated derivative (m1ΨTP) are crucial components in mRNA vaccine technology. Here, an atom‐economic, high‐yielding biocatalytic route for ΨTP synthesis is presented. A new chemoenzymatic route for m1ΨTP synthesis was developed.
