Controlling Release Kinetics of an Adjuvant from a Depot Improves the Efficacy of Local Immunotherapy in Metastatic Cancer

open access: yesAdvanced Science, EarlyView.
We show that intratumoral release of an immunotherapy—CpG —can be tuned with molecular precision by complexing it to an elastin‐like polypeptide (ELP). Injecting ELP‐CpG and an excipient ELP into tumors drives thermally‐triggered phase separation into a micro‐scale “sponge” depot that releases CpG at rates depending on the ELP composition, which ...

Bacterial Foreignization Nanosystem Elicits Multi‐Phenotypic T Cells for Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
In this study, a comprehensive strategy for tumor immunogenicity reshaping is implemented, in which all adjuvanticity, antigenicity, and reactogenicity of tumor cells are highly improved by an engineered bacterial nanosystem through a selective membrane fusion process. The resulting foreignized tumor cells evoke potent innate immune responses, inducing

Brachyury‐Activated Fucoidan Hydrogel Microspheres Rejuvenate Degenerative Intervertebral Discs Microenvironment

open access: yesAdvanced Science, EarlyView.
This study describes the creation of injectable Brachyury (Bry) mRNA‐loaded Fucoidan‐functionalized GelMA hydrogel microspheres (BLNP@GF). These microspheres efficiently suppress inflammation, enhance extracellular matrix (ECM) synthesis in degenerated nucleus pulposus cells, and mitigate intervertebral disc degeneration (IVDD) in rat models.

One-month evaluation of safety and immunogenicity of a Vi-DT typhoid conjugate vaccine (Bio-TCV®) in Indonesian subjects 6 months to 45 years: A phase III randomized clinical trial

open access: yesVaccine: X
Background: Typhoid fever typically affects children and young adults and is a considerable cause of morbidity and mortality. Vaccination is the most effective preventive strategy to control typhoid fever in the short to medium term.
