Targeting the MDM2‐MDM4 interaction interface reveals an otherwise therapeutically active wild‐type p53 in colorectal cancer

open access: yesMolecular Oncology, EarlyView.
This study investigates an alternative approach to reactivating the oncosuppressor p53 in cancer. A short peptide targeting the association of the two p53 inhibitors, MDM2 and MDM4, induces an otherwise therapeutically active p53 with unique features that promote cell death and potentially reduce toxicity towards proliferating nontumor cells.

Open Continual Feature Selection via Granular-Ball Knowledge Transfer [PDF]

open access: yesarXiv
This paper presents a novel framework for continual feature selection (CFS) in data preprocessing, particularly in the context of an open and dynamic environment where unknown classes may emerge. CFS encounters two primary challenges: the discovery of unknown knowledge and the transfer of known knowledge.

Highly multiplexed digital PCR assay for simultaneous quantification of variant allele frequencies and copy number alterations of KRAS and GNAS in pancreatic cancer precursors

open access: yesMolecular Oncology, EarlyView.
Combining melting curve analysis enhances the multiplexing capability of digital PCR. Here, we developed a 14‐plex assay to simultaneously measure single nucleotide mutations and amplifications of KRAS and GNAS, which are common driver genes in pancreatic cancer precursors. This assay accurately quantified variant allele frequencies in clinical samples

Exploring Feature-based Knowledge Distillation for Recommender System: A Frequency Perspective [PDF]

open access: yesarXiv
In this paper, we analyze the feature-based knowledge distillation for recommendation from the frequency perspective. By defining knowledge as different frequency components of the features, we theoretically demonstrate that regular feature-based knowledge distillation is equivalent to equally minimizing losses on all knowledge and further analyze how ...

A large‐scale retrospective study in metastatic breast cancer patients using circulating tumour DNA and machine learning to predict treatment outcome and progression‐free survival

open access: yesMolecular Oncology, EarlyView.
There is an unmet need in metastatic breast cancer patients to monitor therapy response in real time. In this study, we show how a noninvasive and affordable strategy based on sequencing of plasma samples with longitudinal tracking of tumour fraction paired with a statistical model provides valuable information on treatment response in advance of the ...

Prioritized base Debugging in Description Logics [PDF]

open access: yes, 2015
International audienceThe problem investigated is the identification within an input knowledge base of axioms which should be preferably discarded (or amended) in order to restore consistency, coherence, or get rid of undesired consequences.

Bring Your Own Knowledge: A Survey of Methods for LLM Knowledge Expansion [PDF]

open access: yesarXiv
Adapting large language models (LLMs) to new and diverse knowledge is essential for their lasting effectiveness in real-world applications. This survey provides an overview of state-of-the-art methods for expanding the knowledge of LLMs, focusing on integrating various knowledge types, including factual information, domain expertise, language ...

ShcD adaptor protein drives invasion of triple negative breast cancer cells by aberrant activation of EGFR signaling

open access: yesMolecular Oncology, EarlyView.
We identified adaptor protein ShcD as upregulated in triple‐negative breast cancer and found its expression to be correlated with reduced patient survival and increased invasion in cell models. Using a proteomic screen, we identified novel ShcD binding partners involved in EGFR signaling pathways.

Targeting the AKT/mTOR pathway attenuates the metastatic potential of colorectal carcinoma circulating tumor cells in a murine xenotransplantation model

open access: yesMolecular Oncology, EarlyView.
Dual targeting of AKT and mTOR using MK2206 and RAD001 reduces tumor burden in an intracardiac colon cancer circulating tumor cell xenotransplantation model. Analysis of AKT isoform‐specific knockdowns in CTC‐MCC‐41 reveals differentially regulated proteins and phospho‐proteins by liquid chromatography coupled mass spectrometry. Circulating tumor cells
