ROWVA: A Structure‐Based Metric for Predicting the Pathogenicity of Protein Variants Using Alphafold2

This study investigates whether several AF2‐derived scores can predict functional scores and pathogenicity labels, and suggests that the root mean square deviation between the wild‐type and variant structures predicted by AlphaFold2 (ROWVA) may be the most effective predictor. This approach may also be applicable to other multimeric proteins.
