Highly Efficient Generation of Germline Mutations Using CRISPR/Cas9 in the Speckled Wood Butterfly Pararge aegeria

We demonstrate the efficient use of CRISPR/Cas9 to generate germline loss‐of‐function (LOF) mutations in the speckled wood butterfly (Pararge aegeria), an ecological model species. Targeting the yellow‐y gene, we achieved nearly 80% mosaic LOF phenotypes in adults and high germline mutation rates in offspring.
