Survival outcomes and treatment patterns in myelofibrosis in the JAK inhibitor era

open access: yesBritish Journal of Haematology, EarlyView.
Summary Data on real‐world survival and treatment patterns in large, unselected myelofibrosis (MF) cohorts remain limited. We analysed 1649 patients with MF diagnosed from 2010 onwards and included in the Spanish Myelofibrosis Registry across 64 centres. Janus kinase (JAK) inhibitors were administered to approximately half of the patients.

Treatment sequence with gilteritinib and allogeneic haematopoietic stem cell transplantation in relapsed/refractory FMS‐like tyrosine kinase 3 (FLT3)‐mutated acute myeloid leukaemia patients: A multicentre real‐world study

open access: yesBritish Journal of Haematology, EarlyView.
Summary Patients with relapsed or refractory acute myeloid leukaemia (r/r AML) harbouring FMS‐like tyrosine kinase 3 (FLT3) mutations generally have poor prognosis and limited treatment options. The second‐generation FLT3 inhibitor gilteritinib provides better disease control compared to standard relapse therapies and was approved based on two ...

Salvage treatment patterns in multiple myeloma patients progressing after lenalidomide maintenance: A real‐life study from European Myeloma Network (EMN) Italy

open access: yesBritish Journal of Haematology, EarlyView.
Summary The extensive use of lenalidomide (Len) in the first‐line treatment of multiple myeloma (MM) has resulted in an increased frequency of patients who have been exposed or are refractory to Len (Len‐R) at first relapse. However, the outcome of Len‐R patients treated at first relapse after autologous haematopoietic stem cell transplantation (AHSCT)

Minimal residual disease–guided ibrutinib and venetoclax in patients with chronic lymphocytic leukaemia and complex karyotype

open access: yesBritish Journal of Haematology, EarlyView.
Minimal residual disease (MRD)–guided ibrutinib and venetoclax (IVen) achieved durable remissions and high undetectable MRD (uMRD) rates in patients with high‐risk chronic lymphocytic leukaemia (CLL). Compared with historical ibrutinib monotherapy, IVen was associated with improved progression‐free and overall survival despite presence of complex ...

Distinct clinical and genetic characteristics of myelodysplastic syndrome in younger patients

open access: yesBritish Journal of Haematology, EarlyView.
Summary Myelodysplastic neoplasms (formerly myelodysplastic syndromes, MDS) are heterogeneous clonal haematological malignancies that primarily affect the elderly, though a notable proportion of patients are diagnosed at younger ages. We retrospectively analysed 1437 patients diagnosed or treated at Asan Medical Center between 1989 and 2022, comparing ...

Real‐world outcomes of ide‐cel and cilta‐cel in relapsed/refractory multiple myeloma: A nationwide, retrospective analysis in Austria

open access: yesBritish Journal of Haematology, EarlyView.
Summary B‐cell maturation antigen‐directed chimeric antigen receptor (CAR) T cells have revolutionized the treatment of relapsed/refractory multiple myeloma. However, no randomized head‐to‐head comparison of idecabtagene vicleucel (ide‐cel) and ciltacabtagene autoleucel (cilta‐cel) is available, and real‐world data suggest differences in efficacy and ...

Predicted microbiome‐associated metabolic pathways are linked to immune checkpoint modulation in chronic lymphocytic leukaemia

open access: yesBritish Journal of Haematology, EarlyView.
Summary Microbial metabolites are key regulators of host immunity, yet their functional impact in chronic lymphocytic leukaemia (CLL) remains unclear. Following up on our earlier observations of microbiome dysbiosis in CLL, we performed predictive metabolic profiling based on 16S ribosomal RNA (rRNA) sequencing of oral and stool samples collected from ...

Functional assessment of inherited myeloid neoplasm‐associated SAMD9L germline variants via Monoallelic CRISPR modelling

open access: yesBritish Journal of Haematology, EarlyView.
Summary While the majority of myeloid neoplasms are sporadic, the increasing application of germline genetic testing has led the World Health Organization to designate ‘Myeloid malignancies with germline predisposition’ as a distinct clinical entity, carrying major implications for clinical care and research.
