Purification and characterization of thiol dependent, oxidation-stable serine alkaline protease from thermophilic Bacillus sp.

Alkaline serine protease was purified to homogeneity from culture supernatant of a thermophilic, alkaliphilic Bacillus sp. by 80% ammonium sulphate precipitation followed by CM-cellulose and DEAE-cellulose ion exchange column chromatography.

Purification of Rennet by DEAE-Cellulose Chromatography

レンネットをDEAE-セルロースカラムに吸着させ, 0.1M燐酸緩衝液(pH 5.7)に0.1, 0.2および0.3M NaClを加えて調製した溶出液および0.1N HClで順次溶出させてレンニンを分離精製し,あわせてレンネット中に混在するペプシンをも分離し,その活性割合を測定した. (1) 0.1M燐酸緩衝液(pH 5.7)に添加するNaClの濃度を0.1Mから0.2Mに増加した溶出液で得られるFraction 3に凝乳力カが存在し, pH 6.62,イオン強度0.2の燐酸緩衝液を用いた電気泳動,沈降分析からみて均一性の高いレンニンが得られた.

Purification of peptidyl-tRNA on benzoylated DEAE-cellulose columns

The o-nitrophenylsulfenyl group, and amino protecting group in the chemical synthesis of peptidyl-tRNA was used to attach newly synthesized peptidyl-tRNA to a benzoylated DEAE-cellulose column. This facilitated the isolation of a highly purified specific tRNA with a well defined peptide chain.
