Deep mutational scanning for therapeutic antibody engineering

Trends in Pharmacological Sciences, 2022
The biophysical and functional properties of monoclonal antibody (mAb) drug candidates are often improved by protein engineering methods to increase the probability of clinical efficacy. One emerging method is deep mutational scanning (DMS) which combines the power of exhaustive protein mutagenesis and functional screening with deep sequencing and ...

Intragenic compensation through the lens of deep mutational scanning

Biophysical Reviews, 2022
A significant fraction of mutations in proteins are deleterious and result in adverse consequences for protein function, stability, or interaction with other molecules. Intragenic compensation is a specific case of positive epistasis when a neutral missense mutation cancels effect of a deleterious mutation in the same protein.

Biological fitness landscapes by deep mutational scanning

2020
Knowledge of the distribution of fitness effects (DFE) of mutations is critical to the understanding of protein evolution. Here, we describe methods for large-scale, systematic measurements of the DFE using growth competition and deep mutational scanning.

Engineering Proteins by Combining Deep Mutational Scanning and Yeast Display

2022
Protein engineering using display platforms such as yeast display and phage display has allowed discovery of proteins with therapeutic and industrial applications. Antibodies and T cell receptors developed for therapeutic applications are often engineered by constructing libraries of mutations in loops of five to ten residues called complementarity ...

Application of Deep Mutational Scanning in Hepatitis C Virus

2018
Mutagenesis is one of the key techniques in virus research. The recent development of deep mutational scanning allows the assessment of replication fitness effects of a large number of viral mutants in a high-throughput manner. Here, we describe a protocol for studying hepatitis C virus (HCV) using deep mutational scanning, which includes the ...
