Novel (S)‐5′‐guanidinopropyl uridine analogs bearing 2′‐O‐methyl or 2′‐fluoro substitutions enhance nuclease resistance and mismatch discrimination while preserving A‐form RNA structure and potent RNAi activity. These modifications provide a promising strategy for improving the stability, specificity, and therapeutic potential of siRNA‐based medicines.

RNA structural profiling of Turnip Yellow Mosaic Virus by DMS‐MaPseq and DREEM analyses uncover that viral genome‐wide RSS is highly complicated and heterogeneous, with alternative RSSs widely distributed across the genome. Notably, the viral 3’ tRNA‐like structure adopts alternative conformations in vivo.

2'‐O‐Acetal Levulinic Ester Ribonucleoside 3'‐Phosphoramidites for the Solid‐Phase Synthesis of Long RNA

Abstract Solid‐phase RNA synthesis is challenging because the 2'‐hydroxyl requires a protecting group that is stable during chain assembly yet easily removable without degrading the product. Driven by the demand for long RNA therapeutics, we report a synthesis method using 2'‐O‐acetal levulinic ester (ALE) ribonucleoside 3'‐O‐phosphoramidites.
