Prevalent and dynamic binding of the cell cycle checkpoint kinase Rad53 to gene promoters

open access: yeseLife, 2022
Replication of the genome must be coordinated with gene transcription and cellular metabolism, especially following replication stress in the presence of limiting deoxyribonucleotides.

Metabolic Origin, Role and Fate of the Denaturant Guanidine. [PDF]

open access: yesMicrob Biotechnol
The origin of metabolic guanidine is largely a mystery. We suggest it is created when guanine‐containing nucleotides are oxidised by molecular oxygen instead of being broken down into urea as purines normally would. Guanidine may act as a signal to help cells control the level of reactive oxygen species.

Optimization of Chemical Defined Medium for Lactobacillus brevis Based on Artificial Intelligence. [PDF]

open access: yesEng Life Sci
ABSTRACT Lactobacillus brevis (L. brevis) is a bacterium known for its lactic acid production and probiotic properties. However, the limited research on industrial fermentation of L. brevis and its low productivity pose challenges. This work aimed to develop a cost‐effective synthetic medium as an alternative to a complex medium for its large‐scale ...

A combination of a DNA-chimera siRNA against PLK-1 and zoledronic acid suppresses the growth of malignant mesothelioma cells in vitro. [PDF]

open access: yes, 2010
Although novel agents effective against malignant mesothelioma (MM) have been developed, the prognosis of patients with MM is still poor. We generated a DNA-chimeric siRNA against polo-like kinase-1 (PLK-1), which was more stable in human serum than the ...

RRM2B: An oxygen-requiring protein with a role in hypoxia

open access: yesMolecular & Cellular Oncology, 2017
How tumor cells adapt and survive under hypoxia significantly impacts patient prognosis. We recently demonstrated that the oxygen-requiring ribonucleotide reductase (RNR) enzyme, which provides cells with deoxyribonucleotides, responds to limited oxygen ...

Deoxycytidine kinase augments ATM-Mediated DNA repair and contributes to radiation resistance. [PDF]

open access: yesPLoS ONE, 2014
Efficient and adequate generation of deoxyribonucleotides is critical to successful DNA repair. We show that ataxia telangiectasia mutated (ATM) integrates the DNA damage response with DNA metabolism by regulating the salvage of deoxyribonucleosides ...

Pseudomonas aeruginosa Nonphosphorylated AlgR Induces Ribonucleotide Reductase Expression under Oxidative Stress Infectious Conditions

open access: yesmSystems, 2023
Ribonucleotide reductases (RNRs) are key enzymes which catalyze the synthesis of deoxyribonucleotides, the monomers needed for DNA replication and repair.

The cyanobacterial ribosomal-associated protein LrtA from Synechocystis sp. PCC 6803 is an oligomeric protein in solution with chameleonic sequence properties [PDF]

open access: yes, 2018
The LrtA protein of Synechocystis sp. PCC 6803 intervenes in cyanobacterial post-stress survival and in stabilizing 70S ribosomal particles. It belongs to the hibernating promoting factor (HPF) family of proteins, involved in protein synthesis. In this

Unbalanced Deoxyribonucleotide Synthesis Caused by Methotrexate [PDF]

open access: yesBMJ, 1972
The effect of methotrexate on the free intracellular pools of thymidylate triphosphate (dTTP) and deoxyadenosine triphosphate (dATP) in normal human phytohaemagglutinin-transformed lymphocytes has been studied. Methotrexate caused a fall in the dTTP pool ranging from 38% to 88% and a rise in the dATP pool ranging from 24% to 185%.A rise in the free ...

MTBSTFA derivatization-LC-MS/MS approach for the quantitative analysis of endogenous nucleotides in human colorectal carcinoma cells

open access: yesJournal of Pharmaceutical Analysis, 2022
Endogenous ribonucleotides (RNs) and deoxyribonucleotides (dRNs) are important metabolites related to the pathogenesis of many diseases. In light of their physiological and pathological significances, a novel and sensitive pre-column derivatization ...
