ABSTRACT Immune checkpoint inhibitors (ICIs) frequently cause thyroid immune‐related adverse events (irAEs). Permanent hypothyroidism, the clinically most relevant phenotype, requires lifelong hormone replacement. We investigated its incidence, course, risk factors, and prognostic relevance under pembrolizumab.

A Pan‐Cancer Single‐Cell Atlas of Bone Metastases Reveals Convergent Malignant Programs and Coordinated Immune Dysfunction

A pan‐cancer atlas deciphers tumor‐induced reprogramming in bone metastases. Malignant evolution toward MYC/proliferation is a hallmark of bone metastases. Niche‐mediated immune dysfunction is characterized by immature pDCs, impaired antigen presentation and phagocytosis in TAMs, and exhausted T cells.
