Pyruvate dehydrogenase operates as an intramolecular nitroxyl generator during macrophage metabolic reprogramming

open access: yesNature Communications, 2023
M1 macrophages enter a glycolytic state when endogenous nitric oxide (NO) reprograms mitochondrial metabolism by limiting aconitase 2 and pyruvate dehydrogenase (PDH) activity. Here, we provide evidence that NO targets the PDH complex by using lipoate to

Kinetic Model for a Threshold Filter in an Enzymatic System for Bioanalytical and Biocomputing Applications [PDF]

open access: yesJ. Phys. Chem. B 118, 12435-12443 (2014), 2014
A recently experimentally observed biochemical "threshold filtering" mechanism by processes catalyzed by the enzyme malate dehydrogenase is explained in terms of a model that incorporates an unusual mechanism of inhibition of this enzyme that has a reversible mechanism of action.

Differences in gene expression of enzymes involved in branched-chain amino acid metabolism of abdominal subcutaneous adipose tissue between pregnant women with and without PCOS

open access: yesTaiwanese Journal of Obstetrics & Gynecology, 2021
Objective: Polycystic ovary syndrome (PCOS) appears to be a common endocrine disorder of women in reproductive age. Adipose tissue (AT) is known as an active tissue in the metabolism of branched-chain amino acids (BCAA; Valine, Leucine, and Isoleucine ...

GC-MS Profile of Diodella sarmentosa (SW) Bacigalupo El Cabral ex Borhidi Ethanol Leaf Extract and its Total Dehydrogenase Inhibitory Potential [PDF]

open access: yesarXiv, 2021
Phytochemical composition of ethanol leaf extract of Diodella sarmentosa was profiled with GC-MS and the inhibitory property of the extract against total microbial dehydrogenases were assessed. The major constituents of the extract were squalene (29.50%), Phytol (24.68%), phenol, 3-pentadecyl- (18.58%), 1-Butanol, 3-methyl- (9.09%) and n-Hexadecanoic ...

Purification and characterization of acetoin:2,6-dichlorophenolindophenol oxidoreductase, dihydrolipoamide dehydrogenase, and dihydrolipoamide acetyltransferase of the Pelobacter carbinolicus acetoin dehydrogenase enzyme system [PDF]

open access: yesJournal of Bacteriology, 1991
Dihydrolipoamide dehydrogenase (DHLDH), dihydrolipoamide acetyltransferase (DHLTA), and acetoin: 2,6-dichlorophenolindophenol oxidoreductase (Ao:DCPIP OR) were purified from acetoin-grown cells of Pelobacter carbinolicus. DHLDH had a native Mr of 110,000, consisted of two identical subunits of Mr 54,000, and reacted only with NAD(H) as a coenzyme.

Transcriptional and Proteomic Analysis of a Ferric Uptake Regulator (Fur) Mutant of Shewanella oneidensis: Possible Involvement of Fur in Energy Metabolism, Transcriptional Regulation, and Oxidative Stress [PDF]

open access: yes, 2002
The iron-directed, coordinate regulation of genes depends on the fur (ferric uptake regulator) gene product, which acts as an iron-responsive, transcriptional repressor protein. To investigate the biological function of a fur homolog in the dissimilatory

Collaborative learning of images and geometrics for predicting isocitrate dehydrogenase status of glioma [PDF]

open access: yesarXiv, 2022
The isocitrate dehydrogenase (IDH) gene mutation status is an important biomarker for glioma patients. The gold standard of IDH mutation detection requires tumour tissue obtained via invasive approaches and is usually expensive. Recent advancement in radiogenomics provides a non-invasive approach for predicting IDH mutation based on MRI.

Evaluation of Shifts of Gene Transcription Levels of Unicellular Green Alga Chlamydomonas reinhardtii Due to UV-C Irradiation

open access: yesMicroorganisms, 2023
Green algae produce valuable lipids as carbon-recycling resources. Collecting whole cells with the intracellular lipids could be efficient without cell burst; however, direct use of the cells causes microbial contamination in environments.

Folding of the Protein Domain hbSBD [PDF]

open access: yesBiophysical J. 89, 3353 (2005), 2006
The folding of the alpha-helice domain hbSBD of the mammalian mitochondrial branched-chain alpha-ketoacid dehydrogenase (BCKD) complex is studied by the circular dichroism technique in absence of urea. Thermal denaturation is used to evaluate various thermodynamic parameters defining the equilibrium unfolding, which is well described by the two-state ...
