DNA Synthesis in vitro [PDF]

open access: possibleNature, 1970
In a new in vitro system, DNA is synthesized semiconservatively at rates of chain growth comparable with replication in vivo. This DNA synthesis is also observed with a strain of E. coli, which lacks DNA polymerase activity in vitro.

Viral DNA Synthesis [PDF]

open access: possible, 1991
In 1973, it was first discovered that human hepatitis B virus (HBV) preparations from sera of chronically infected individuals contain an endogenous polymerase activity capable of carrying out a repair reaction on the viral DNA template in the presence of magnesium and nucleotide triphosphates (Kaplan et al. 1973).

Translesion DNA Synthesis and Reinitiation of DNA Synthesis in Chemotherapy Resistance

Biochemistry (Moscow), 2020
Many chemotherapy drugs block tumor cell division by damaging DNA. DNA polymerases eta (Pol η), iota (Pol ι), kappa (Pol κ), REV1 of the Y-family and zeta (Pol ζ) of the B-family efficiently incorporate nucleotides opposite a number of DNA lesions during translesion DNA synthesis.

The elemental role of iron in DNA synthesis and repair.

Metallomics, 2017
Iron is an essential redox element that functions as a cofactor in many metabolic pathways. Critical enzymes in DNA metabolism, including multiple DNA repair enzymes (helicases, nucleases, glycosylases, demethylases) and ribonucleotide reductase, use ...

DNA synthesis and myogenesis

Experimental Cell Research, 1961
Abstract Differentiating muscle cells synthesizing myosin, the meromyosins, and actin do not concurrently synthesize DNA. Presumptive myoblasts which synthesize DNA do not concurrently synthesize myosin, the meromyosins or actin. The multinucleated skeletal muscle fiber is the product of cell fusion.

The Synthesis of Designer DNA

2018
The synthesis of designer DNA requires an approach where the user can determine both the sequence and the number of nucleobases. The protocol outlined here describes an enzymatic method for the synthesis of long repeat-sequence DNA. The method utilizes a PCR-based approach; starting with short oligo-seeds, c.a. 20 bp, bearing a minimum of two repeating

DNA Functional Materials Assembled from Branched DNA: Design, Synthesis, and Applications.

Chemical Reviews, 2020
DNA is traditionally known as a central genetic biomolecule in living systems. From an alternative perspective, DNA is a versatile molecular building-block for the construction of functional materials, in particular biomaterials, due to its intrinsic ...

Cloning of senescent cell-derived inhibitors of DNA synthesis using an expression screen.

Experimental Cell Research, 1994
We here describe a rapid and simple expression screen method that has allowed us to isolate cDNAs coding for inhibitors of DNA synthesis from senescent human diploid fibroblasts.

DNA Synthesis Security

2012
It is generally assumed that genetic engineering advances will, inevitably, facilitate the misapplication of biotechnology toward the production of biological weapons. Unexpectedly, however, some of these very advances in the areas of DNA synthesis and sequencing may enable the implementation of automated and nonintrusive safeguards to avert the ...
