Replication independent formation of extrachromosomal circular DNA in mammalian cell-free system. [PDF]

open access: yesPLoS ONE, 2009
Extrachromosomal circular DNA (eccDNA) is a pool of circular double stranded DNA molecules found in all eukaryotic cells and composed of repeated chromosomal sequences. It was proposed to be involved in genomic instability, aging and alternative telomere

The landscape of extrachromosomal circular DNA (eccDNA) in the normal hematopoiesis and leukemia evolution

open access: yesCell Death Discovery, 2022
Elevated extrachromosomal circular DNA (eccDNA) has been reported to accelerate tumor pathogenesis. Although the eccDNA profiles of other tumors have been established, the landscape of the eccDNA of acute myeloid leukemia (AML) has not been revealed. Our

Identification and characterization of eccDNA-driven genes in humans

open access: goldPLOS One
Extrachromosomal circular DNA (eccDNA) amplification promotes oncogene expression and cancer development. However, the global transcriptional landscape mediated by eccDNA has not yet been extensively profiled. Here we report a comprehensive analysis spanning cancer, non-cancerous disease and health by developing a new approach to catalog eccDNA-driven ...

Analysis of multiple‐herbicide resistant Amaranthus palmeri populations from Spain points to an introduction of the eccDNA from America

open access: hybridPest Management Science, EarlyView.
In this study, multiple herbicide resistant mechanisms have been confirmed in two Amaranthus palmeri populations from Spain that survived glyphosate‐ and ALS‐inhibitor herbicides. Our data suggest that the eccDNA and the mutations identified in the ALS gene are the result of an evolutionary event that occurred outside Spain.

EccDNA‐Driven VPS41 Amplification Alleviates Genotoxic Stress via Lysosomal KAI1 Degradation (Adv. Sci. 25/2025)

open access: goldAdvanced Science, Volume 12, Issue 25, July 3, 2025.
Extrachromosomal Circular DNA In article number 2501934, Bin Shi, Shuyu Zhang, and co‐workers present the increased existence of eccDNA induced by ionizing radiation. VPS41 gene in the eccDNA confers protection against genotoxic stress. This study highlights the role of eccDNA in cellular defense, providing strategies to enhance tissue resilience to ...

scEccDNAdb: an integrated single-cell eccDNA resource for human and mouse

open access: yesDatabase
Extrachromosomal circular DNA (eccDNA), an extrachromosomal circular structured DNA, is extensively found in eukaryotes. Exploring eccDNA at the single-cell level contributes to understanding heterogeneity, evolution, development, and specific functions ...

A comprehensive atlas of full-length Arabidopsis eccDNA populations identifies their genomic origins and epigenetic regulation

open access: goldPLoS Biology
Extrachromosomal circular DNA (eccDNA) has been described in several eukaryotic species and has been shown to impact phenomena as diverse as cancer and herbicide tolerance. EccDNA is thought to arise mainly through transposable element (TE) mobilization.

Autonomous replication sequences from the Amaranthus palmeri eccDNA replicon enable replication in yeast [PDF]

open access: greenBMC Research Notes, 2020
Abstract Objective: The objective of the research presented here was to determine whether autonomous replication sequences (ARS) discovered in the eccDNA replicon of glyphosate resistant Amaranthus palmeri enable self-replication in a yeast system.

Investigating cellular heterogeneity at the single-cell level by the flexible and mobile extrachromosomal circular DNA [PDF]

open access: yesComputational and Structural Biotechnology Journal, 2023
Extrachromosomal circular DNA (eccDNA) is a special class of DNA derived from linear chromosomes. It coexists independently with linear chromosomes in the nucleus.
