The presence of tumor extrachomosomal circular DNA (ecDNA) as a component of liquid biopsy in blood
Medical Hypotheses, 2018In molecular biology covalently closed circular DNAs are able to passthrough double layer of eukaryotic cellular membrane. Very recently the presence of circular extra chromosomal DNA (ecDNA) has been shown which are different in seventeen different types of cancers. In fact, ecDNA are the tricky way of oncogenes to increase their copy number.

Extrachromosomal circular DNA (ecDNA) have been found in most types of human cancers, and ecDNA incorporating viral genomes has recently been described, specifically in human papillomavirus (HPV)-mediated oropharyngeal cancer (OPC).

Abstract IA017: Extrachromosomal DNA (ecDNA): Cancer’s dynamic circular genome
Cancer ResearchHuman genes are arranged on 23 pairs of chromosomes, but in cancer, tumor-promoting genes and regulatory elements can free themselves from chromosomes and relocate to circular, extrachromosomal pieces of DNA (ecDNA).

Tumors exhibit dynamic gradients of oxygen diffusion and consumption leading to regions of hypoxia. The presence of hypoxia is associated with poor clinical prognosis and elevated genomic instability. Genes can be amplified extrachromosomally as ecDNA,
