Simulation of the regulation of EGFR endocytosis and EGFR‐ERK signaling by endophilin‐mediated RhoA‐EGFR crosstalk [PDF]
Deregulations of EGFR endocytosis in EGFR‐ERK signaling are known to cause cancers and developmental disorders. Mutations that impaired c‐Cbl–EGFR association delay EGFR endocytosis and produce higher mitogenic signals in lung cancer. ROCK, an effector of small GTPase RhoA was shown to negatively regulate EGFR endocytosis via endophilin A1.

Osimertinib or Platinum–Pemetrexed in EGFR T790M–Positive Lung Cancer
BACKGROUND Osimertinib is an epidermal growth factor receptor tyrosine kinase inhibitor (EGFR‐TKI) that is selective for both EGFR‐TKI sensitizing and T790M resistance mutations in patients with non–small‐cell lung cancer.

Brk/PTK6 sustains activated EGFR signaling through inhibiting EGFR degradation and transactivating EGFR [PDF]
Epidermal growth factor receptor (EGFR)-mediated cell signaling is critical for mammary epithelial cell growth and survival; however, targeting EGFR has shown no or only minimal therapeutic benefit in patients with breast cancer. Here, we report a novel regulatory mechanism of EGFR signaling that may explain the low response rates. We found that breast

Non-small-cell lung cancer (NSCLC) constitutes 85% of patients diagnosed with lung cancer. In metastatic cases, its treatment classically consists of systemic cytotoxic chemotherapy, which resulted in a median overall survival of 7.9 months.

Optimized EGFR Blockade Strategies in EGFR Addicted Gastroesophageal Adenocarcinomas [PDF]
Abstract Purpose: Gastric and gastroesophageal adenocarcinomas represent the third leading cause of cancer mortality worldwide. Despite significant therapeutic improvement, the outcome of patients with advanced gastroesophageal adenocarcinoma is poor. Randomized clinical trials failed to show a

Resistance mechanisms to osimertinib in EGFR-mutated non-small cell lung cancer
Osimertinib is an irreversible, third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that is highly selective for EGFR-activating mutations as well as the EGFR T790M mutation in patients with advanced non-small cell lung ...
