Abstract Protoplasts were enzymatically prepared from the mesocarp of two species of oil palm (Elaeis guineensis Jacq. and E. oleifera HBK and Cortes) 16–20 weeks after anthesis and from rapidly multiplying embryogenic cultures of E. guineensis. The protoplasts were purified by density gradient centrifugation in 20% (w/v) sucrose.

Dura is the main variety found in natural groves, and has been the source of palm oil for decades – well before modern methods of oil palm cultivation were introduced to Africa in the second quarter of the twentieth century.

The oil palm, Elaeis guineensis, (Fig. 1) is a monocotyledonous tree belonging to the coccoid group of palms. It grows over a period of years to a height of up to 15 m over a lifetime of 100 years or more. Its productive commercial life is between 20 and 30 years, after which harvesting becomes increasingly difficult, and yields decline slowly.
