Adaptive Network Stabilization Despite Profound Structural Reorganization in Human Organotypic Cultures

open access: yesAdvanced Science, EarlyView.
Adult human organotypic cortical slices undergo marked neuronal loss, laminar disruption, and glial activation, yet their network activity progressively stabilizes after a variable period. Electrophysiology, longitudinal calcium imaging, and quantitative histology show that structurally remodeled human circuits show adaptive capacity resulting in ...

Hederagenin Attenuates Morphine Addiction via GABAA1‐Mediated Modulation of Ca2+–CaMK–CREB Signaling in Ventral Tegmental Dopaminergic Neurons

open access: yesAdvanced Science, EarlyView.
Hederagenin is identified as a GABAA1‐targeting natural compound that suppresses morphine‐associated reward by restraining Ca2+–CaMK–cAMP–CREB signaling in ventral tegmental dopaminergic neurons. RVG29‐modified liposomal delivery further enhances brain accumulation and behavioral efficacy, providing a mechanistically defined strategy for natural ...

Printing‐Assisted Integration of Thermal, Fluidic and Electrical Components in Implants for Focal Brain Cooling

open access: yesAdvanced Science, EarlyView.
Extrusion‐based 3D printing of silicones and discrete electronic components are combined to create cortical implants for rodents that integrate thermal neuromodulation and electrical recording. Linked to an autonomous auxiliary driver unit, the system enables rapid, stable and localized hypothermia alongside ECoG recording.

A Gut‐Brain Axis Underlying Succinate‐Associated Compulsive‐Like Behavior

open access: yesAdvanced Science, EarlyView.
Gut‐derived succinate is associated with an excitatory‐inhibitory imbalance in dmPFC glutamatergic neurons, involving microglial SUCNR1 activation and vagal afferent signaling via upregulated gut 5‐HT4 receptors along the DVC‐CeA‐dmPFC circuit, changes that are linked to excessive compulsive‐like behavior.

Temporal‐Targeted Accelerated 40 Hz tACS Enhances Cognitive Function and Network Integration in Alzheimer's Disease: A Randomized Clinical Trial

open access: yesAdvanced Science, EarlyView.
Bilateral temporal accelerated 40‐Hz tACS improves cognitive function in patients with Alzheimer's disease. Resting‐state fNIRS identifies treatment‐related increases in frontotemporoparietal functional connectivity, which are associated with cognitive improvement. ABSTRACT The temporal lobes are key hubs for memory and cognition.

The Deubiquitinase UCHL1 Drives Susceptibility to Atrial Fibrillation by Stabilizing CaMKII‐δ

open access: yesAdvanced Science, EarlyView.
UCHL1 is highly upregulated in mice and patients with AF. UCHL1 directly binds to and removes K48‐linked polyubiquitin chains from CaMKII‐δ at K251 and enhances CaMKII‐δ stability, which activates RyR2, disrupts Ca2+ handling, and promotes AF. These findings establish UCHL1 as a critical regulator of CaMKII‐δ stability and AF‐related Ca2+ handling ...

Brain Network Dynamics of Local and Global Predictive Processing in Aging

open access: yesAdvanced Science, EarlyView.
Separation of concurrent whole‐brain networks in source‐reconstructed magnetoencephalography (MEG) data suggests that healthy aging reorganizes, rather than uniformly attenuates, neural responses elicited from hierarchical auditory violations. Enhanced early sensory deviance processing alongside reduced higher‐order cognitive responses suggests a large‐
