Time to take human embryo culture seriously.

open access: yesHuman Reproduction, 2016
STUDY QUESTION Is it important that end-users know the composition of human embryo culture media? SUMMARY ANSWER We argue that there is as strong case for full transparency concerning the composition of embryo culture media intended for human use ...

On subcellular distribution of the zinc finger 469 protein (ZNF469) and observed discrepancy in the localization of endogenous and overexpressed ZNF469

open access: yesFEBS Open Bio, EarlyView.
ZNF469 regulates the expression of genes encoding extracellular matrix proteins. Endogenous ZNF469 is predominantly cytoplasmic, while in transfected cells, it forms aggregates reminiscent of biomolecular condensates, located mainly in the nucleus. These condensates exhibit overlapping staining with proteasomes and are also associated with the mitotic ...

Deep learning mediated single time-point image-based prediction of embryo developmental outcome at the cleavage stage [PDF]

open access: yesarXiv, 2020
In conventional clinical in-vitro fertilization practices embryos are transferred either at the cleavage or blastocyst stages of development. Cleavage stage transfers, particularly, are beneficial for patients with relatively poor prognosis and at fertility centers in resource-limited settings where there is a higher chance of developmental failure in ...

Co‐expression of HSV‐1 ICP34.5 enhances the expression of gene delivered by self‐amplifying RNA and mitigates its immunogenicity

open access: yesFEBS Open Bio, EarlyView.
ICP34.5 is one of the most important antihost response proteins. The saRNA‐encoding HSV‐1 neurovirulence protein ICP34.5 clearly mediated the eukaryotic initiation factor 2 alpha subunit (eIF2α) dephosphorylation and significant suppression of innate immune responses in vitro, leading to enhanced expression of the saRNA‐encoded gene.

Apolipoprotein CIII regulates lipoprotein-associated phospholipase A2 expression via the MAPK and NFκB pathways

open access: yesBiology Open, 2015
Apolipoprotein CIII (apo CIII), a small glycoprotein that binds to the surfaces of certain lipoproteins, is associated with inflammatory and atherogenic responses in vascular cells.

Atlas-based automated detection of swim bladder in Medaka embryo [PDF]

open access: yesarXiv, 2019
Fish embryo models are increasingly being used both for the assessment of chemicals efficacy and potential toxicity. This article proposes a methodology to automatically detect the swim bladder on 2D images of Medaka fish embryos seen either in dorsal view or in lateral view. After embryo segmentation and for each studied orientation, the method builds

Matrigel inhibits elongation and drives endoderm differentiation in aggregates of mouse embryonic stem cells

open access: yesFEBS Open Bio, EarlyView.
Stem cell‐based embryo models (SCBEMs) are valuable to study early developmental milestones. Matrigel, a basement membrane matrix, is a critical substrate used in various SCBEM protocols, but its role in driving stem cell lineage commitment is not clearly defined.

A unique regulatory phase of DNA methylation in the early mammalian embryo

open access: yesNature, 2012
DNA methylation is highly dynamic during mammalian embryogenesis. It is broadly accepted that the paternal genome is actively depleted of 5-methylcytosine at fertilization, followed by passive loss that reaches a minimum at the blastocyst stage. However,

Hsa-miR-30d, secreted by the human endometrium, is taken up by the pre-implantation embryo and might modify its transcriptome

open access: yesDevelopment, 2015
During embryo implantation, the blastocyst interacts with and regulates the endometrium, and endometrial fluid secreted by the endometrial epithelium nurtures the embryo.

Prostaglandin F2α (PGF2α) production possibility and its receptors expression in the early- and late-cleaved preimplantation bovine embryos

open access: yesBMC Veterinary Research, 2019
Background Prostaglandin F2α (PGF2α) is an important component for the physiology of female reproductive processes. In the literature, the data pertaining to the synthesis and action of PGF2α in early embryonic bovine development are limited.
