Combination vs. single‐drug nonprescription analgesics for acute pain management: A narrative review

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Combining nonprescription analgesics with different mechanisms of action has been proposed as a rational strategy to optimize the management of acute pain. This review assessed the efficacy and safety of nonprescription analgesics, including paracetamol (acetaminophen), metamizole and nonsteroidal anti‐inflammatory drugs (NSAIDs) used in combination vs.

Effect of vepdegestrant, a PROTAC oestrogen receptor degrader, on dabigatran and rosuvastatin pharmacokinetics in healthy participants

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims To evaluate the effects of vepdegestrant, a PROteolysis TArgeting Chimera (PROTAC) oestrogen receptor degrader, on the pharmacokinetics of dabigatran (a P‐glycoprotein [P‐gp] substrate) and rosuvastatin (a breast cancer resistance protein [BCRP] substrate).

Recurrent fexuprazan‐induced liver injury following unintentional re‐exposure

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Fexuprazan, a potassium‐competitive acid blocker, is increasingly used for the treatment of gastroesophageal reflux disease in South Korea. While generally well tolerated, data regarding its hepatotoxic potential are scarce. We report a case of idiosyncratic drug‐induced liver injury in a 61‐year‐old woman following 2 months of fexuprazan therapy.

Enhancing patient stratification methods for medication review during acute admissions

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Medication review in the hospital is an effective tool for identifying inappropriate prescribing, but not every patient can receive one due to resource constraints. Thus, it is important to identify patients who stand to benefit most from medication review.

Effectiveness and safety of combining thiazides with loop diuretics vs. loop diuretics monotherapy in patients with acute decompensated heart failure: A retrospective cohort study

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims This study investigated factors associated with and outcomes of combined loop diuretics with hydrochlorothiazide (HCTZ) for acute decompensated heart failure (ADHF). Methods In this retrospective study, adults hospitalized for ADHF and who received intravenous loop diuretics between January 2016 and March 2023 were enrolled.

AZD0449, an inhaled JAK1 inhibitor, in healthy participants and patients with mild asthma

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims Inhaled corticosteroids (ICS) plus bronchodilator are recommended for the treatment of asthma. Targeting the JAK1‐dependent pathway may be an alternative for asthma management in patients with incomplete response to ICS. The aim of this study was to investigate the safety, tolerability, pharmacokinetics (PK) and pharmacodynamics (PD) of AZD0449, a
